聚苹果酸微胶囊在药物缓释中的应用

论文研究了以聚苹果酸和壳聚糖为囊材,以硝苯地平为囊芯,采用复凝聚(complex coacervation)法制备微胶囊。利用P-B实验设计对复凝聚体系形成条件进行了初步的筛选
摘要:β-聚苹果酸是(β-poly malic acid,PMLA)一种水溶性脂肪族聚酯,且具有生物相容性和生物可吸收性,其降解产物无毒性(半数致死量LD/50=3.3g/kg,腹膜内),无抗原性,也可以被人体吸收利用,因此可作为药物载体和胶囊材料。本毕业论文研究了以聚苹果酸和壳聚糖为囊材,以硝苯地平为囊芯,采用复凝聚(complex coacervation)法制备微胶囊。利用P-B实验设计对复凝聚体系形成条件进行了初步的筛选,在此基础上,利用正交实验设计深入研究了影响微胶囊粒径和分散系数的各种因素,获得适宜的制备工艺条件,即聚苹果酸的浓度为1.0g/L、壳聚糖的浓度为0.5g/L、聚苹果酸与壳聚糖溶液的体积比为2:1、壳聚糖的pH值为4.0、硝苯地平溶液的浓度为1.0g/L、硝苯地平的添加量为3.0mL、聚苹果酸溶液的滴加速度为5.0mL/h、搅拌速度为600r/min、体系反应时间为45min,所得微胶囊的粒径为130.7nm,分散系数为0.304,包埋率达到43.0%。17201

Poly-microcapsule in the Application of Drug Release
Abstract: Poly ( malic acid ) is a kind of water soluble aliphatic polyester, and a biocompatible and biodegradable, its degradation product without toxicity (LD/50=33g/kg, intraperitoneal ), no antigenicity, can also be absorbed by the body, therefore can be used as drug carrier and capsule material. This study is based on chitosan and poly (malic acid) as wall material, with complex coacervation method for nifedipine were entrapped. Using the P-B experiment design screed the preliminary formation conditions in complex condensed system, on this basis, study the various factors of the impact of the microcapsule particle size and dispersion coefficients by using the orthogonal experimental design in-depth. At the last, we would obtain the suitable preparation conditions, it is that the poly malic acid concentration is 1.0g/L, the concentration of chitosan for 0.5g/L, poly malic acid and chitosan solution volume ratio of 2:1, chitosan, pH value of 4,nifedipine solution concentration is 1.0g/L, nifedipine added to 3.0mL, poly malic acid solution droplets acceleration is 5.0mL/h, stirring speed is 600r/min, reaction time is 45min, the particle size of the microcapsules for 130.7nm,  PDI is 0.304, the embedding rate reached 43%.
