油菜的F-BOX蛋白序列特征与生物信息分析

摘要:F-BOX蛋白普遍存在于真核生物体中, 具有 F-BOX结构域的蛋白家族 ,在泛素介导的蛋白质类物质水解的过程中,具有底物识别特性的功能,在细胞凋亡、细胞周期调控、细胞信号传导、转录调控等生命过程中具有重要的作用。从NCBI数据库中查询下载了油菜F-BOX蛋白序列,利用生物信息学技术,对其基本理化性质、电子表达、基因结构和保守域、蛋白紊乱区和球蛋白区、二级结构和三级结构等方面进行分析和预测。结果表明,油菜F-BOX蛋白序列中20种氨基酸含量差异较大;该蛋白为球形亲水蛋白,无跨膜区,N端不含信号肽,为非分泌蛋白;其基因有五个外显子,主要在花和种子中表达;二级结构以无规则卷曲为主;用Swiss-Model进行建模,该蛋白是α类蛋白,Ramachandran 图检测表明该模型结构符合立体化学规则。本文为进一步研究F-BOX蛋白的结构及其功能提供了理论基础。32130

Sequence characters and Bioinformatics Analysis of F-BOX Protein in a Brassica napus L
Abstract:F-BOX protein is a kind of protein family with F-BOX structure domain widely existing in eukaryotic organism,which has the function of the substrate recognition characteristics in the process of the protein ubiquitin mediated hydrolysis,  and plays an important role in apoptosis,cell cycle regulation, cell signal transduction, transcription regulation and so on.Downloaded from the NCBI database F-BOX protein sequence of Brassica napus L, using bioinformatics technology, the expression of the basic physical and chemical properties, electronic, gene structure and conservative, disorder of protein domain area and globulin area, secondary structure and tertiary structure is analyzed and forecasted.The results showed that the content of 20 kinds of amino acids in F-BOX protein sequence of Brassica napus L is unbalanced;The protein is spherical hydrophilic protein, no transmembrane region, excluding signal peptide protein N end and is not a secreted protein;The protein gene have five exons, which is mainly expressed in flowers and seeds;Secondary structure is given priority to with random curl;The protein is the alpha protein with Swiss-Model for modeling,and Ramachandran map test showed that the Model structure comply with the rules of stereochemistry.This paper provides a theoretical basis for further studying the F-BOX protein structure and function .
