摘要:透明质酸是一种大分子粘多糖,其生物活性和使用效果具有Mr相关性,应根据使用目的的不同,选择合适的分子量。近年来不同相对分子量透明质酸功能及应用的研究进展,其中详细介绍了高分子量的HA具有较好的粘弹性、保湿性、抑制炎性反应、润滑、药物缓释等功能,及在骨关节炎的治疗、术后防粘连、眼科手术黏弹剂和滴眼液等领域的广泛应用。低分子量HA(LMWHA)和HA寡聚糖(o-HA)有抗肿瘤、促进伤口愈合、促进血管生成、免疫调节等作用,且通过对HA的降解可获得LMWHA和o-HA 。
透明质酸的功能决定了其应用的范围。而透明质酸的功能又与其分子量大小密切相关,即不同相对分子质量的透明质酸具有不同的生物活性和使用效果。本次实验主要对不同分子量(100KD和180KD)透明质酸在注射水体系下的主要理化特性进行研究,对浓度分别为0.05%、0.1%、0.3%和0.5%的高、低分子量透明质酸溶液先运用Brookfield粘度计进行粘度测定、再在43%以及81%的干燥环境中进行吸水性和保水性的测定、最后运用AR-G2流变仪器测取稳态和动态流变。结果表明:不同分子量的透明质酸水溶液中,5℃环境下浓度为0.5%的水溶液粘度较佳,且0.5%高分子量透明质酸水溶液较0.5%低分子量透明质酸水溶液粘度更大;吸水性与保水性是在放置96h后浓度为0.5%的水溶液最强;稳态流变性随剪切率增加而降低,因此浓度为0.05%的透明质酸水溶液稳态流变最佳,而动态流变性随角频率增加而增加,则0.5%浓度的透明质酸水溶液动态流变性最佳。17523

Abstract:Hyaluronic acid (HA) is a type of macromolecule glycosidically, its biological activity and using effect are related to the Mr relativity, the suitable molecular weight should be chosen by the different intended use. Recently, many researches are done to the function and the application of different molecular weight hyaluronic acid, and introducing the HA of high molecular weight has better viscoelasticity、water keeping、restrain inflammatory reaction、lubrication、drug controlled-release and etc. in detail.Also widespread use in the treatment in osteoarthritis、adhesion preventing after operation、stick the agent and eye drops in ophthalmology operation and many other areas. Low molecular weight HA(LMWHA)and oligosaccharide have the effect of anti-tumor、promoting wound healing and angiogenesis、immune adjustment and etc., and the LMWHA and the 0-HA can be gotten through the HA degradation.
The function of HA determines its applied scope. And the HA function is closely related to its molecular weight as well, it means the HA of different molecular weight have different biological activity and the using effect. The experiment mainly studied in  the performance of different molecular weight hyaluronic acid in injection water system,  The solution concentration of 0.05%、0.1%、0.3% and 0.5% high molecular weight and low molecular weight have been put into the experiment.Firstly using the Brookfield viscometer determined the viscosity. Then testing the hygroscopicity and the water-retaining property under the dry environment of 43% and 81%. Finally using the AR-G2 rtheology determined rtheology in steady state and frequency state.And the results showed that among the different molecular weight HA,the 0.5% HA in 5 ℃ had the better rtheology, and the rtheology of 0.5% high molecular weight HA was better than the low molecular weight HA of 0.5%;The better hygroscopy and the water-retaining property was the 0.5% water solution after placed 96 hours;The rtheology behavior in steady state was reduced with the shear rate increased,so the 0.05% HA had the better steady state,while the frequency state was increased with the angular frequency increased, and the better frequency state  was the 0.5% HA. 不同分子量透明质酸在注射水体系中性能研究:http://www.lwfree.cn/yixue/20180111/19209.html
